Rota Rod is the instrument meant for studying the effect of diazepam on the muscle – grip strength of mice.

 

A versatile instrument built with LSI Chips is equipped with three compartments and three independent 4 digits Digital milliseconds timer for measuring the fall-off time of the mice.

 

It is also equipped with a motor coupled with rotating rod whose speed can be varied with the help of the front panel knob marked with an arrow or speed less than 10 RPM.

Speed is marked from 5 RPM to 35 RPM.

 

One of the important pharmacological actions of antianxiety agents of benzodiazepine class of drugs is muscle relaxing property. The skeletal muscle relaxation together with taming or calming effect these agents reduce anxiety and tension. The loss of muscle-grip is an indication of muscle relaxation. This can be easily checked in animals using Rota Rod.

 

The difference in the fall off time from the rotating rod between the control and diazepam treated animal is taken as index of muscle relaxation. The rate of rotation of the rod should be adjusted such that a normal mouse can stay on the rod for an appropriate period of time.

 

OPERATION

  • Before switching ON the instrument, ensure that the platforms in which the mice has to fall are laying in the backward direction. This platform is basically a SEA-SAW adjustment such that when animal or mice fell down from the rotating rod, this platform also imitacion bolso goyard pressed in the forward direction thereby stops the timer and the time is recorded in the timer until it is resetted.

  • After this switch ON the instrument. There are chances that random reading may appear in the digital timer displays.

  • Now RESET the timers with the help of the PUSH SWITCHES provided below the TIME DISPLAY.

  • After all this adjustments, press the PUSH SWITCH marked START.
  • The motor starts rotating thereby rotating the rod mounted above the compartments. Adjust the speed of the motor with the help of the control knob with an arrow/speed as per requirement of the experiment. 20-25 rpm is an ideal speed.
  • Now your instrument is ready for experimentation.
  • Weigh the animals and number them.

  • Now switch OFF the instrument again and ensure again that the platforms are lying in the backward directions.
  • Switch ON the instrument once and see that the motor is not running and all the timers are stopped.
  • In case the timers are registering the time and are running then start pressing the platforms one by one  in front direction so that they are made to reset on the ELECTRONIC SWITCH and this will STOP the timers & also the motor gets stopped automatically.
  •  Place the animal on the rotating rod . In your instrument, you have the privileage of placing two animals at a time as your instrument is having two compartments.
  • Bring back the platforms in their original position i.e. lying in the backward   direction and not in contact with the ELECTRONIC SWITCH.
  • Now press the START push switch once and with this action the motor starts rotating and timer starts running.
  • Note down the “FALL OFF TIME” when the mouse falls from the rotating rods
  •  This can be read from the timer which was stopped by the fall of the mouse.
  • Inject diazepam to all  the animals. After 30 minutes repeat the experiment as above and note the fall off time.
  • Compare the fall off time of animals before and after diazepam treatment.

  • Do not handle with wet hands.
